Job Description

AbbVie Inc is embracing new paradigms and technologies to develop novel and life-changing therapies across a diverse range of therapeutic areas. The Quantitative, Translational ADME Sciences (QTAS) department at AbbVie is seeking a highly motivated and energetic bioanalytical scientist to join the biologics characterization and bioanalysis group. The main function of this position will be to develop and implement LC-MS (liquid chromatography-mass spectrometry)-based bioanalysis and in vitro characterization assays of biologics entities, including monoclonal antibody, multi-specific antibody, antibody drug conjugate (ADC), lipid nanoparticles (LNP), Adeno-Associated Virus (AAV) etc. The ideal candidate will be experienced in the optimization and implementation of novel and routine LC-MS-based bioanalytical methods to support in-vivo PK and TK studies as well as in-vitro stability characterization of biologics and genetic medicines. The candidate will be responsible for routine sample preparation, method development, instrument maintenance, data analysis, writing reports, and presenting data at internal and external meetings.


Key tasks and responsibilities:


  • Optimize and implement bioanalytical methods for quantifying therapeutic drugs (including but not limited to antibodies, ADCs, fusion proteins, LNPs, and small molecules) in biological matrices using LC-MS MRM assay.
  • Perform high-throughput sample preparation in both biofluids and tissues to support both PK/PD and proteomics studies.
  • Support biologics and genetic medicine drug development by developing in vitro characterization assays using high resolution MS systems.
  • Routinely operate LC-MS and automation instruments, including maintenance and troubleshooting.
  • Write and maintain protocols and facilitate assay optimization and implementation to ensure data quality.
  • Utilize and maintain electronic laboratory notebooks and LIMS systems for study documentation.
  • Communicate data interpretation internally and externally.

Qualifications

  • Degree in chemistry, pharmacology or other related fields. BS with 8+ years in a pharmaceutical industry setting with a focus on mass spectrometry.
  • Highly motivated individual with a strong intellectual and technical background in bioanalytical and characterization methods development, specifically for antibodies, LNP, AAV and oligonucleotide using LC-MS/MS. Must have direct experience in both small and large molecule quantitation (in biological matrices), peptide mapping, PTM analysis, intact/reduced protein analysis (in biological matrices) and lipid profiling in LNPs.
  • Deep and broad knowledge in separation science and mass spectrometry. Extensive hands-on experience in maintaining and troubleshooting SCIEX Triple Quad b5500/6500/6500+, Agilent QTOF 6545/6550 and Thermo Orbitrap mass spectrometers is required.
  • Experience in performing immunoaffinity purification and sample preparation using automation systems (including but not limited to AssayMap Bravo and TECAN) is a must.
  • Experience in using LIMS system for sample and data management is a must.
  • Strong data analytical skills are required. Experience with Skyline, Protein Metrics, Analyst, Xcalibur and MassHunter is preferred.
  • Knowledge in drug discovery and related disciplines. In-depth understanding in vivo PK, TK studies and corresponding bioanalytical assays is preferred.
  • Excellent verbal and written communication skills, strong analytical and organizational abilities, and the ability to work independently and thrive in a team environment.
  • Desire to excel in a fast-paced environment, with strong problem-solving skills, and the ability to prioritize tasks and meet deadlines effectively.
